고지방식이를 섭취한 마우스에서 양파껍질 열수 추출물이 혈중지질에 미치는 영향

Effects of Onion Peel Water Extract on the Blood Lipid Profiles in Mice Fed a High-Fat Diet

Onion (Allium cepa L.) is one of the richest sources of flavonoids in human diet. Onion peel contains over 20 times more quercetin than onion flesh. In this study, we studied the effects of onion peel water extract (OPE) on the blood lipid profiles in mice. The onion peel extracts was extracted with hot water. The experimental groups were divided with 3 groups (n = 6) of ICR male mice: normal diet + distilled water (NC), high-fat diet + distilled water (HF), high-fat diet + onion peel water extract 20 ㎎/㎏ (OPE-20). The oral administration was conducted daily. The experimental period was 7 weeks. Onion peel water extract showed higher concentration of polyphenol gallic acid and anti-oxidant trolox equivalent than the ethanol extract. The body weight gain and food efficiency ratio was significantly lower in the OPE-20 group as compared with HF group (p < 0.05). The epididymal fat and retroperitoneal fat showed significantly lower weights and sizes in the OPE-20 group as compared with HF group (p < 0.05). The serum levels of total cholesterol, LDL cholesterol and triglyceride were significantly lower in the OPE-20 group as compared with HF group (p < 0.05). The OPE-20 group showed higher HDL cholesterol concentration than HF group (p < 0.05). Atherogenic index was ignificantly lower in as compared with HF group (p < 0.05). The serum levels of glucose, GOT and GPT were significantly lower in the OPE-20 group as compared with HF group (p < 0.05). In these results, we suggests that onion peel water extracts supplementation can reduces the serum lipid components and improves the lipid metabolism in hyperlipidemic mice induced with a high-fat diet.

서 언

혈중 지질의 증가는 동맥경화 및 비만과 같은 심각한 질병 들과 밀접한 관련이 있기 때문에 혈중 지질 조절에 도움을 줄 수 있는 천연물 소재의 개발에 대한 연구 필요성이 크게 대두 되고 있는 실정이다 (Lim and Choi, 2001; Yoon et al., 2010). 양파 (Allium cepa L.)는 한약재로서 옥총(玉葱), 양총 (洋葱)이라고 하며 위장의 소화력을 도와주고 장관의 작용을 활성화 시킨다 (Ra et al., 1997; Woo et al., 2003).). 양파는 식재료 및 향신 조미료로 가장 많이 사용되는 식품 중 하나로 서 quercitrin, rutin과 같은 flavonoid (Miean and Mohamed, 2001)와 황 화합물인 allyl propyldisulfide 및 diallyl disulfide 와 같은 phytochemical이 함유되어 있어 (Ra et al., 1997), 다양한 생리적 기능성을 지니는 것으로 알려져 있다. 양파의 수분은 약 88-90%, 탄수화물은 6.8-10.0%로 조성으로는 과당 (fructose)이 가장 많고, 포도당 (glucose)과 설탕 (sucrose)은 거의 같은 양이 포함되어 있으며 다당류도 많다. 이들 다당류 를 구성하는 당은 galactose, arabinose, ribose 및 rhamnose 등이 있으며 전분은 거의 함유되어 있지 않다 (Ra et al., 1997).

양파의 주요한 생리활성물질로는 강력한 항산화능과 체 내 지방 수준을 감소시켜주는 기능을 가진 플라보노이드 (flavonoid)와 황 화합물 (sulphur compounds)이 대표적이다 (Jaime et al., 2001; Woo et al., 2003). Miean과 Mohamed (2001)의 연구에 따르면 양파는 62가지 열대 식물 중 flavonoid 함량이 가장 높다고 보고되었으며 보통 폐기물로 인 식되어지는 건조된 양파껍질에서는 효과적인 항산화제로 작용 하는 플라보노이드 배당체 등이 있는 것으로 알려져 있다 (Brahma et al., 2009).

양파 껍질은 양파에 비하여 약 10-100배의 flavonoids를 함 유하며, 주요한 flavonoid는 quercetin이다 (Bang and Cho, 1998). Quercetin은 폴리페놀 화합물인 플라보노이드의 일종으 로 식품을 통해 섭취하는 폴리페놀 중 양적으로 가장 중요하 고 과일이나 채소 등에 주로 존재하며 (Brahma et al., 2009) 주로 당과 결합하여 quercetin glucoside 형태로 많이 존재하 고 (Wach et al., 2007) quercetin-4'-glucoside와 quercetin- 3,4'-diglucoside가 전체 flavonoid의 80%를 차지할 정도로 가 장 대표적인 형태이다 (Bonaccorsi et al., 2008).

Quercetin은 in vivo 및 in vitro 연구에서 다양한 약리효과 가 규명되어 왔다 (Chen et al., 2004). Quercetin은 free radical을 소거하여 t-butyl hydroperoxide와 같은 유해물질에 의해 발생하는 돌연변이를 억제하는 기전이 보고되었으며 흡 연으로 인해 발생하는 스트레스에 관여하는 단백질의 생성도 억제하는 것으로 알려져 있다 (Edenharder and Grunhage, 2003; Pinot et al., 1997). Quercetin은 순환기계에도 강력한 약리작용을 발휘하는데, 심근세포 세포손상 억제, 동맥경화방 지작용 등이 알려져 있으며 혈관 평활근을 이완시켜서 혈압을 낮추거나 부정맥을 억제하기도 한다 (Psotova et al., 2004; Daniel et al., 2003; Duarte et al., 2001; Soloviev et al., 2002). 또한 UV에 의한 피부손상의 억제, 항당뇨병작용 등 산 화적 스트레스가 관여하는 다른 질환에 대해 유효하다 (Vessal et al., 2003; Mahesh and Menon., 2004). 또한 뇌에서 산화 를 일으키는 물질인 peroxynitrate의 함량을 낮추는 효과가 있 는 것으로 알려져 있다 (Shutenko et al., 1991).

최근 양파의 발암 물질 활성 감소 능력 및 항암 효과에 대 해서도 다수의 연구가 진행되었다 (Jang and Lim, 2009). 양 파에서 flavonoids 같은 polyphenolic compounds가 항산화적 작용이 강하고 항암작용 기전에 기여하는 바가 크다는 것 을 증명해 주고 있으며, 이는 양파의 quercetin과 그의 glycosylated derivatives의 화학적 예방작용을 기대하게 한다고 보고되었다 (Siess et al., 1997). Alkyl sulphides와 diallyl disulphids 역시 발암물질의 대사에 관여하는 효소를 변화시키 면서 암화과정 초기에서 예방적 작용하는 것으로 보고되었다 (Siess et al., 1997).

지금까지 양파에 대한 실험은 양파 육질에 국한되어 있었고, 양파 가공 시 약 10%가 껍질 부분으로 폐기되거나 퇴비로 사 용되고 있다는 점을 고려할 때, 식품의 가식부로 이용되지 못 하던 양파 껍질에서 생리적 유용성과 건강 기능 식품으로의 활용 가능성을 확인하는 것은 경제적으로도 큰 가치를 지닌다 고 할 수 있다. 이러한 양파 껍질을 이용한 연구는 폐자원을 활용한다는 면에서 의미가 있다 (Joo et al., 1991; Chung et al., 2011).

따라서 본 연구에서는 건강기능성 소재로서 인식이 높은 양 파껍질의 열수 추출물을 통한 혈중지질 개선에 미치는 효과를 검증하고자 하였다.


재료 및 방법
1. 재료 및 추출

본 실험에 사용된 양파는 재배되어 수확된 양파의 껍질을 수집하여 3회 세척과정을 거쳐 암소에서 건조 한 후 건식분쇄 기로 분쇄하여 분말을 시료로 사용하였다. 건조된 양파껍질 분 말을 정제수를 이용하여 20㎎/mL의 농도로 만든 후 100°C, pH 6에서 30분간 열수추출하고 Rotary Evaporator (BUCHI Rotavapor R-220, Flawil, Switzerland)를 이용하여 65°C에서 20시간 감압농축한 후 동결건조하여 실험에 사용하였다. 열수 추출물과의 비교를 위한 에탄올추출물 역시 같은 조건에서 추 출되었다.

2. 총 플라보노이드 측정

열수 추출액 및 에탄올 추출액을 95% 에탄올에 희석하여, 95% 에탄올, 증류수, 10% Al(NO3)39H2O, 1M CH3COOK 및 증류수를 차례로 가하여 충분히 교반한 이후 실온에서 40 분간 방치한 이후에 415㎚에서 Spectrophotometer (Mecasys, Daejeon, Korea)로 흡광도를 측정하였다.

3. 폴리페놀 측정

열수 추출액 및 에탄올 추출액을 95% 에탄올에 희석하여, Folin-Ciocalteau Reagent (Sigma, MO, USA), 증류수을 차례 로 가한 이후 교반한 이후 상온에 2분간 방치. 그리고 15% Na2CO3, 증류수를 차례로 가하고 충분히 교반한 이후 실온 에서 120분 정치한 이후에 755㎚에서 Spectrophotometer (Mecasys, Daejeon, Korea)로 흡광도를 측정하였다.

4. 동물실험 군구성 및 식이

본 실험에서는 평균 체중이 33.53 ± 1.87 g인 13주령의 수컷 ICR mouse를 Samtako (Osan, Korea)에서 구입하여 사용하 였다. 1주간 기본사료로 적응시킨 후, 일반식이군 (NC), 고지 방식이군 (HF), 양파껍질 중농도 고지방식이군 (OPE-20) 3군 으로 분류하여 군당 6마리씩 7주간 실험하였고, 양파껍질 중 농도 고지방식이군 (OPE-20)은 매일 경구투여 하였다. 사료와 음수는 자유롭게 섭취하도록 하였다. 동물실험은 원광대학교 동물실험 윤리위원회의 지침에 준하여 수행하였다 (Approval No. WKU 13-42). 사료는 NC 군에게는 일반 마우스 배합사 료 (Samtako, Osan, Korea)를 급여하고, HF와 OPE-20군에는 High Fat Diet 60% (Saeronbio Inc., Uiwang, Korea)와 필 터 및 자외선 살균기로 여과 살균된 정제수를 자유롭게 섭취 하도록 하였다. 사용된 사료의 조성은 Table 1과 같았다.

5. 부검 및 시료채취

고지혈증 유발 및 시료처리 7주 후 실험동물을 12시간 절 식을 유지시킨 뒤 diethyl ether를 이용하여 호흡마취 시키고, 복대정맥에서 채혈하고 혈청을 분리한 후 −80°C에 동결보존 하였다. 병리조직학적 검사를 위해 간 (liver) 및 지방조직 (Epididymal fat, Retroperitoneal fat, Peri-renal fat)을 적출하 여 무게를 측정하였다.

6. 혈액 생화학적 분석

혈중 지질은 총 콜레스테롤 (TC), 중성지방 (TG), 고밀도 지단백 콜레스테롤 (HDL), GOT (Glutamic Oxaloacetic Transaminase), GPT (glutamic yuvic transaminase)와 혈당 (glucose)은 아산제약 kit를 사용하여 자동생화학검사기 BS-220 (Mindray, Shenzhen, China)로 측정하였다. 측정된 총 콜레스 테롤, HDL 콜레스테롤, 중성지방을 이용해 저밀도 지단백 콜 레스테롤 (LDL), 동맥경화지수 (Atherogenic index, AI), 심장 위험지수 (Cardiac risk factor, CRF)를 계산하였다.

7. 병리조직학적 검사

간 조직 및 지방조직은 (epididymal fat, retroperitoneal fat, peri-renal fat) 10% 중성 포르말린에 고정하고, 병리조직학적 검사를 위한 통상적인 방법을 사용하여 파라핀 포매한 후, 4µm 두께로 절편을 하고 슬라이드 제작한 후 일반적인 Hematoxylin & Eosin (H&E) 염색을 실시하고 Nikon Eclipse E200 (Nikon, Tokyo, Japan) 현미경을 이용하여 NASH (nonalcoholic steatohepatitis) score를 측정하였다 (Neuschwander- Tetri and Caldwell, 2003).

8. 통 계

실험을 통하여 얻어진 모든 결과는 평균±표준편차 (mean ± SD)로 나타내었다. 각 시험군의 유의성에 대한 통계 는 대조군과 실험군 간의 통계학적 비교를 위해 One-way ANOVA SPSS v. 12 (SPSS INC., Chicago, USA)를 사용 하여 분석하였으며, Duncan’s Multiple Range Test (DMRT) 검정을 실시하여 (p < 0.05) 이하일 때 통계적으로 유의한 것 으로 판단하였다.


결과 및 고찰
1. 양파껍질 열수 및 에탄올 추출물의 생리활성 측정 결과

본 연구에서는 유기용매 추출물이 아닌 열수추출물을 시료 로 사용하였는데, 이는 추출 시 제기되는 잔류용매의 독성문 제를 피하여 화학 합성 항균제 및 보존제의 대체효과와 식물 자체에 포함되어 있는 다양한 생리활성 물질 섭취효과를 동시 에 기대 할 수 있으며, 아울러 향후 기능성 차 또는 건강기능 식품 소재로 개발시 경제적 이점이 있을 것으로 사료되었기 때문이다 (Eloff, 1998). 본 실험에서 열수추출물과 에탄올 추 출물과 비교하였을 경우 총 폴리페놀과 항산화물질은 증가한 반면 총 플라보노이드는 감소하였다 (Table 2). 이러한 결과를 통하여 다양한 생리활성을 고려하였을 때 열수추출물이 효율 적이라고 사료 된다.

2. 체중 변화 및 사료 섭취량

체중변화는 7주간 관찰 기간 동안의 체중 증감에 있어 4주 부터 6주까지 정상군이 고지방식이군에 비해 유의적인 차이가 없었다 (Fig. 1). 실험 종류 후, 군간 체중 증가와 사료섭취량 을 체중과 비교하여 분석한 결과, 체중증체와 사료효율은 고 지방식이 급여군인 HF군의 마우스 보다 양파껍질 열수 추출 물 투여군인 OPE-20군의 마우스가 유의하게 낮은 것을 확인 할 수 있었다 (p < 0.05) (Table 3). Chung (2011) 등의 양파 추출물 용량별 투여에 따른 체중 및 식이 효율에서 초기 체중 은 6군 간의 유의적인 차이가 없었다. 실험 기간인 8주 동안 223.2-241.3 g의 체중 증가를 보였으며, 실험기간 동안의 체중 증가량은 6군에서 유의적인 차이를 보이지 않은 것으로 나타 났다.

3. 장기중량 변화

간의 절대장기 무게와 상대장기 무게는 각 실험군 간의 유 의적인 차이를 보이지 않았으나 OPE-20군이 더 낮은 경향을 보였다 (Table 4).

부고환지방과 후복막지방의 무게는 고지방식이 급여군인 HF 군의 마우스 보다 양파껍질 열수 추출물 투여군인 OPE-20군 의 마우스가 유의하게 낮은 것을 확인할 수 있었다 (p < 0.05) (Table 4). Kim과 Kim 등 (2004)의 연구결과 양파껍질 에탄 올 추출물 섭취 군이 다른 군들에 비해 체중 100 g당 부고 환 지방의 양이 적은 경향을 보여 본 실험 결과와 일치하였다 . 또한 양파껍질 에탄올 추출물이 고지방식이군에 비해 복강 내 지방과 부고환 지방의 중량이 감소하는 경향을 보인 Choi (2009)의 결과와 유사한 것으로 나타났다.

4. 생화학 분석 결과

생화학분석 결과 총 콜레스테롤과 LDL 및 TG의 농도는 정상 NC군에 비해 고지방 식이 HF군에서 유의하게 증가되었 으며, 고지방식이 급여군인 HF군의 마우스 보다 양파껍질 열 수 추출물 투여군인 OPE-20군의 마우스가 유의하게 낮은 것 을 확인할 수 있었다 (p < 0.05) (Table 5). HDL의 농도는 정 상 NC군에 비해 고지방 식이 HF군에서 유의하게 감소되었으 며, 고지방식이 급여군인 HF군의 마우스 보다 양파껍질 열수 추출물 투여군인 OPE-20군의 마우스가 유의하게 높은 것을 확인할 수 있었다 (p < 0.05) (Table 5). 총 콜레스테롤과 HDL 수치를 가지고 계산된 동맥경화지수와 심장위험지수는 정상 NC군에 비해 고지방 식이 HF군에서 유의하게 증가되었 으며, 고지방식이 급여군인 HF군의 마우스 보다 양파껍질 열 수 추출물 투여군인 OPE-20군의 마우스가 유의하게 낮은 것 을 확인할 수 있었다 (p < 0.05) (Table 5). 양파의 지질 강화 효과에 대해서는 선행된 몇몇의 동물실험과 인체시험을 통해 보고된 바 있는데, 예를 들면 당뇨병 유발 동물에게 양파의 공급 시 LDL 콜레스테롤은 감소시키고, HDL 콜레스테롤을 증가시킨다고 발표되었으며 (Campos et al., 2003), 쥐에게 양 파 첨가 청국장을 투여 시 간의 총 지방량과 콜레스테롤 함량 이 대조군에 비해 6.2%, 9.1% 낮았다는 국내 연구 보고가 있 다 (Park et al., 2008).

5. 간손상 지표

양파껍질 추출물의 간손상 지표인 GOT와 GPT 수치에 미 치는 변화는 정상 NC군에 비해 고지방 식이 HF군에서 유의 하게 증가되었으며, 고지방식이 급여군인 HF군의 마우스 보 다 양파껍질 열수 추출물 투여군인 OPE-20군의 마우스가 유 의하게 낮은 것을 확인할 수 있었다 (p < 0.05) (Table 6). 고 지방 식이 또는 고콜레스테롤 식이를 섭취한 경우 간 조직내 중성지방 및 콜레스테롤의 축적을 일으켜 간 무게가 증가 되는데, 과량의 콜레스테롤이 지방 수용체인 아포단백질 (apoprotein)과 결합하여 지단백 (lipoprotein)으로 배출되지 못 하면 지방간 또는 간손상을 유발하게 된다 (Sung et al., 1997). 본 연구에서 양파껍질 열수 추출물은 고지방식이로 유 도된 간손상에 대하여 치료효과가 있는 것으로 확인되었다.

본 연구 결과로 퀘르세틴을 다량 함유하고 있는 양파껍질 열수 추출물 경구투여로 인해 혈중지질은 개선시키고 혈당은 감소시키고 간손상 지표인 GOT와 GPT 감소 효과를 확인하 였다 (Table 6).

7. 병리조직학적 검사

비알코올성 지방간 정상군에 비해 고지방식이군에서 통계적 으로 유의하게 증가하였고 양파껍질 열수 추출물군에서 유의 적으로 감소하였다 (p < 0.05) (Fig. 2).

지방 측정한 결과 부고환지방에서는 정상군에 비해 고지방 식이군에서 통계적으로 유의하게 증가하였고 양파껍질 열수 추출물군에서 유의적으로 감소하였다. 후복막지방에서는 정상 군에 비해 고지방식이군에서 통계적으로 유의하게 증가하였고 양파껍질 열수 추출물군에서 유의적으로 감소하였다 (p < 0.05) (Fig. 3, 4).

자연에서 얻어지는 식물성 식품 속 플라보노이드의 섭취는 많은 역학 연구에서 고혈압, 아테롬성 동맥경화증, 고인슐린 혈증, 비만 등을 예방할 수 있는 것으로 밝혀진 바 있다 (Rivera et al., 2008; Nomura et al., 2008). 대부분의 과일 과 채소에서 흔하게 찾을 수 있는 플라보노이드는 특히 양파, 사과, 포도, 와인, 차 등에 풍부하게 함유되어 있으며, 최근 28 가지 채소와 9가지 과일에서 대표적인 플라보노이드 성분인 quercetin의 양을 측정한 결과 양파에서 그 함량이 가장 풍부 하게 나타났다 (Perez-Vizcaino et al., 2010; Davalos et al., 2006). 특히 양파로부터 얻은 quercetin glycoside는 순수 화합 물과 비교해 생체 내 이용에 있어 더 효과적이라는 연구결과 가 보고되었다 (Hollman et al., 1995).

본 연구 결과 생리활성물질이 다량 함유된 양파껍질 열수 추출물은 혈중내 콜레스테롤을 조절하고 동맥경화 예방에도 효과적이라고 판단되며, 혈당 강하에도 효과가 있는 것으로 확 인 할 수 있었다.
