빌레나무 추출물의 항산화, α-글루코시다아제 억제 및 항미생물 활성

Maesa japonica (Thunb.) is an evergreen shrub belonged to the Myrsinaceae family, which was discovered in 2006 in South Korea. And, its biological functions have not been well studied. In this study, we determined the antioxidant activities, α-glucosidase inhibitory effects and antimicrobial activities of methanol extract and the solvent fractions of M. japonica leaves and twigs. The highest antioxidant activity obtained by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging assay and reducing power assay was found in the ethylacetate fraction of twigs methanol extract, which contained the highest level of total phenolic compounds compared to the other fractions. In addition, ethylacetate fraction of twigs extract exhibited higher inhibitory activities against α-glucosidase (IC50 = 0.8 µg/ml) compared to the IC50 of the buthanol fraction (IC50 = 16 µg/ml) of leaves extract. It showed antimicrobial activities against Bacillus atrophaeus and Bacillus subtilis subsp. Spizizenii. Although the data is too limited, the current study is the first report on biological functions of M. japonica.

서 언

빌레나무 (Maesa japonica (Thunb.) Moritzi & Zoll.)는 빌레나무과에 속하는 상록 덩굴성 소관목으로 국내에는 2006 년에 처음으로 보고되었으며, 빌레나무의 ‘빌레’는 노출된 암 반을 뜻하는 제주 방언으로 토심이 얕은 지역에 주로 자라서 빌레나무로 명명되었다 (Moon et al., 2006). 제주도의 남서쪽 해발 120 ~ 160 미터 부근의 종가시나무 숲속이나 숲 가장자 리에 독립적 혹은 군생으로 분포하며 분포지역이나 개체수가 적어서 희소한 식물로 분류가 된다 (Moon et al., 2006). 주 요 자생지는 중국을 비롯한 대만, 일본 및 베트남의 북부에 분포하는 것으로 알려져 있다 (Yamazaki, 1993; Chen and Pipoly, 1996; Yang, 1999). 지금까지 보고된 제주도내 빌레나 무의 분포는 한경면 저지리나 산양리 등 서부지역 곶자왈 지 대에서 확인되며 곶자왈 내부에 상록활엽수림 또는 2차림의 형성이 양호한 함몰지형에서 주로 나타나는데, 함몰지역인 경 우 일정한 습도가 유지되고 부엽층 형성이 양호하며 종가시 나무, 붉가시나무 등이 분포하는 곳이다 (Song, 2011). 또한, 한라산 남서쪽에서만 자생하여 제주도가 빌레나무의 북한계로 추정이 되고 있다 (Song, 2011).

빌레나무에 대한 연구는 maesaquinone, embelin, rapanone, maesaquinone, acetylmaesaquinone, 2-hydroxy-5-methoxy- 3-pentadecenyl (tridecenyl- and tridecyl-) benzoquinone과 ardisiaquinones 같은 벤조퀴논 유도체나 (Ogawa and Natori, 1965, 1968), maejaposides, maesabalides, maesasaponin과 13,28-epoxy-oleanane saponins 같은 사포닌 성분 함량 (Koike et al., 1999; Foubert et al., 2009)에 대한 보고가 있으며, 계통학적•형태학적 차이에 따른 조사연구 (Caris et al., 2000; Ma and Saunders, 2003; Utteridge and Saunders, 2006) 정도이다.

빌레나무는 대표적인 남방계 식물로 식물지리학적으로 매우 중요한 종이며 곶자왈 지대의 독특한 환경과 결부시켜 연구가 진행되어야 할 것으로 판단된다. 또한 제주도 중산간 이하 지 역의 개발로 인한 개체 수 감소 및 희소성에 따른 무단채취가 예상되며 이에 따른 보존대책도 시급한 실정이다. 본 연구는 지금까지 빌레나무의 구성성분에 대한 함량이나 물질분리, 빌 레나무과의 다른 종이나 속들 간 형태학적 차이에 대한 보고 만 있을 뿐 생리활성에 대한 연구가 전무한 상태여서, 최근 제주도에서 보고된 빌레나무의 생리활성에 대한 연구를 실시 하였으며, 이 자료가 빌레나무에 대한 기초정보가 될 것으로 기대한다.


재료 및 방법
1. 재료

본 연구에 사용한 빌레나무 (Maesa japonica (Thunb.) Moritzi & Zoll.)는 제주시 한경면 저지리에 위치한 저지곶자 왈에서 채취하였으며 김현철 박사로부터 빌레나무에 대한 동 정을 확인 받아 실험하였다. 잎과 가지로 나누어 수돗물에 깨 끗이 씻은 후 음지에서 자연 건조하였으며 믹서기를 이용하여 분말로 만들어서 실험에 이용하였다. 건조분말 100 그램에 메탄올 1리터를 첨가하여 초음파 추출기 (Power sonic 520, Hwashin Co., Yeongcheon, Korea)에 1시간 추출 후 필터링 하였으며 3회 반복하였다. 모아진 메탄올 추출물은 회전농축 기 사용하여 농축하였다. 농축된 메탄올 추출물은 물로 현탁 한 후, 핵산, 에칠아세테이트, 수포화 부탄올을 이용하여 차례 차례 분획을 실시하였다. 각 분획물은 회전농축기를 이용하 여 농축한 후 메탄올 추출물과 각 분획물을 메탄올이나 물 을 이용하여 재용해 후 –20°C에서 보관하면서 분석에 이용하 였다.

2. 총 페놀함량 측정

총 페놀함량은 Folin-Ciocalteu 법에 의하여 측정하였다 (Kim et al., 2010). 메탄올 추출물이나 그 분획물을 2 N Folin-Ciocalteu 시약 50µl와 섞은 후 실온에서 5분간 반응시 켰다. 5분 후, 20% sodium carbonate 0.3ml을 첨가하고 다시 15분간 반응을 시킨 후 증류수 1ml을 첨가하였다. 반응 혼합물의 흡광도는 spectrophotometer (UV-1800, Shimadzu, Tokyo, Japan)를 이용하여 725nm에서 측정하였다. Gallic acid를 표준품으로 하여 표준검량곡선을 작성하였으며, 각 샘 플의 총 페놀함량은 milligram gallic acid 당량 (mg GAE/g extract)으로 표시하였다.

3. 항산화활성 측정

각 샘플의 DPPH 프리라디칼 소거능을 측정하기 위하여, 다 양한 농도의 샘플과 메탄올을 4ml로 맞춘 후 0.15 mM 1,1- diphenyl-2-picryl-hydrazil (DPPH)를 첨가하여 실온의 암소에 서 30분간 반응시켰다. 그 후 반응액을 517nm에서 흡광도를 측정하였으며, DPPH 프리라디칼 소거능은 Hyun 등 (2014)의 방법에 의해 계산하였다. 대조군으로 합성항산화제인 BHT와 천연항산화제인 비타민 E (알파-토코페롤)를 사용하였다.

각 샘플의 환원력은 Hyun 등 (2014)의 방법에 의하여 측정 하였다. 각각의 샘플농도 (100, 200과 300µg/ml)에 0.2 M sodium phosphate buffer (pH 6.6) 0.5ml와 1% potassium ferricyanide 0.5ml를 섞는다. 혼합물은 50°C로 셋팅된 히팅블 럭에서 20분간 반응을 시킨 후 10% trichloroacetic acid 2.5ml를 첨가하여 반응을 정지시켰다. 반응액 (0.5ml)에 동량 의 물과 0.1ml ferric chloride (0.1%, w/v)를 첨가하여 혼합 한 후 700nm에서 흡광도를 측정하였다. 대조군으로 BHT와 비타민 E를 사용하였다.

4. α-Glucosidase 저해활성 측정

각 샘플 50µl에 0.5 U/ml α-glucosidase 효소액 50µl, 200 mM potassium phosphate buffer (pH 6.8) 50µl를 혼 합하여 37°C에서 15분간 배양한 후 3 mM p-nitrophenyl α- D-glucopyranoside (pNPG) 100µl를 첨가하여 37°C에서 10분 간 반응시켰다. 0.1 M sodium carbonate 750µl로 반응을 정지시키고 405nm에서 흡광도를 측정하였다. 샘플 무첨가구 는 negative control로 사용하였으며, 기질 무첨가구는 blank로 사용하였다. α-glucosidase 저해활성은 다음과 같이 계산하 였다.

5. 항미생물 활성 측정

항균 활성의 분석을 위해 5종의 그람양성균; Bacillus atrophaeus (KACC 14742), Kocuria rhizophila (KACC 14744), Micrococcus luteus (KACC 14819), Staphylococcus epidermidis (KACC 14822), Bacillus subtilis subsp. Spizizenii (KACC 14741)와 4종의 그람음성균; Klebsiella pneumonia (KACC 14816), Enterobacter cloacae (KACC 11958), Salmonella enterica subsp. enterica (KACC 10769), Pseudomonas aerµginosa (KACC 10186)을 사용하여 실험하 였다.

모든 균은 한국농업미생물자원센터 (Korean Agricultural Culture Collection, KACC, Suwon, Korea)에서 분양을 받 았다. 항균 활성은 Hyun 등 (2014)의 방법에 따라 임계 희석 법 (critical dilution method)을 이용하여 분석하였다. 빌레나 무 메탄올 추출물 및 분획물의 항균력은 5종의 그람양성균과 4종의 그람음성군에 대하여 측정하였고, 결과는 최소 저해 농 도 (Minimal Inhibitory Concentration; MIC)로 나타냈다.

6. 통계분석

본 연구의 실험 데이터는 평균 ±표준편차로 나타내었다. 실 험군간의 통계학적 분석은 SPSS (Statistical package for social science, Ver 18.0)를 사용하여 one-way analysis of variance (ANOVA) 분석을 실시하였으며 유의적 차이가 있는 항목에 대해서는 Duncan’s Multiple Range Test (DMRT)로 p < 0.05의 수준에서 통계학적 유의성을 검정하였다. 또한, 측 정 변인간의 관련성을 알아보기 위하여 Pearson 상관분석을 실시하였다.


결과 및 고찰
1. 항산화 활성 및 총 페놀 함량 측정

항산화 물질은 심혈관 질환, 동맥 경화증, 관상 동맥 심장 질환, 인슐린 저항성을 포함한 많은 질병 프로세스에 관여하 는 활성산소종 (reactive oxygen species)을 퇴치하여 건강을 유지하도록 도와준다 (Alfadda and Sallam, 2012). Butylated hydroxytoluene (BHT) 및 butylated hydroxyanisole (BHA)는 강력한 합성 항산화제로 알려졌지만, 동물에 대한 독성 및 발암을 일으키는 것으로 보고되고 있다 (Safer and Al- Nughamish, 1999). 따라서 천연 항산화 물질의 기본 소스를 찾 아내는 것이 무엇보다 중요하게 인식되고 있다. 빌레나무 추출 물의 항산화 활성을 조사하기 위하여, 50%의 DPPH 프리라디 칼을 소거하는데 필요한 시료의 농도를 나타내는 RC50 값을 측 정하였다 (Table 1). 잎에서는 부탄올 분획물 (14.0 ± 0.2µg/ml) 이 가장 활성이 높았으며 다음으로 에칠아세테이트 분획물 (14.3 ± 0.1µg/ml), 메탄올 추출물 (23.5 ± 0.7µg/ml), 물 분획물 (38.9 ± 0.0µg/ml) 및 핵산 분획물 (88.9 ± 0.4µg/ml) 순으로 나 타났다. 가지에서는 에칠아세테이트 분획물 (5.8 ± 0.3µg/ml)이 가장 높은 활성을 나타내었으며 다음으로 부탄올 분획물 (12.9 ± 0.4µg/ml), 메탄올 추출물 (37.1 ± 0.5µg/ml), 핵산 분획 물 (109.2 ± 1.3µg/ml) 및 물 분획물 (196.3 ± 5.3µg/ml) 순으 로 활성을 나타내어 잎 추출물과 대조를 이루었다. 전체적으 로 대조구로 이용된 비타민 E보다는 활성이 약하게 나타났 으나, 메탄올 추출물, 에칠아세테이트 분획물 및 부탄올 분획 물은 대조구인 BHT보다 높은 활성을 나타내었다.

메탄올 추출물 및 분획물의 환원력을 측정하였다 (Fig. 1). 이 분석방법에서 하이드록시라디칼은 Fe2+와 H2O2와의 반응에 의해 생성되며, 항산화제는 Fe2+의 킬레이팅에 의해 하이드록 시라디칼의 생성을 감소시킨다 (Bokhari et al., 2013). 잎과 가지 모두에서 샘플의 양이 증가함에 따라 환원력이 증가함을 볼 수 있었다. 에칠아세테이트 분획물에서 환원력이 가장 컸 으며 다음으로 부탄올 분획물, 메탄올 추출물 순이었다. 대조 구로 사용한 BHT나 비타민 E보다는 다소 약한 환원력을 나 타내었다.

식물에서 유래하는 페놀이나 플라보노이드 화합물은 항산화 및 항균 효과와 같은 효능을 나타내기도 한다 (Proestos et al., 2006). 그것은 일반적으로 식물의 산화를 억제하는 능력이 큰 페놀함량과 관련이 깊기 때문이다 (Kim et al., 2010; Hyun et al., 2014). 그러므로 항산화 활성이 메탄올 추출물 및 그 분획물의 페놀함량과 어떤 관련이 있는지를 알아보기 위하여, 각 시료의 총 페놀함량을 분석하였다 (Table 1). 잎의 메탄올 추출물에서 527.6 ± 20.6mg GAE/g을 나타낸 총 페놀함량이 분 획의 과정을 거치면서 에칠아세테이트 분획물 (911.3 ± 12.2mg GAE/g)과 부탄올 분획물 (744.3 ± 2.8mg GAE/g)로 이동함을 알 수 있었다. 반면 물 분획물 (251.2 ± 3.7mg GAE/g) 그리고 핵산 분획물 (93.2 ± 1.4mg GAE/g)에서는 총 페놀함량이 낮 았다. 가지에서는 잎에서와 마찬가지로 에칠아세테이트 분획 물 (1702.0 ± 11.2mg GAE/g), 부탄올 분획물 (673.4 ± 20.6mg GAE/g), 메탄올 추출물 (589.9 ± 24.4mg GAE/g) 순으로 높았 으며 이어서 핵산 분획물 (103.4 ± 4.7mg GAE/g)과 물 분획 물 (80.2 ± 0.9mg GAE/g) 순이었다. 이것은 DPPH 프리라디 칼 소거능과 환원력의 결과와 유사한 경향을 보였으며, 페놀 의 함량이 항산화 능력과 관련이 있음을 나타내었다 (Im and Lee, 2013; Kim et al., 2013).

2. α-Glucosidase 저해 활성 측정

수 많은 약용식물들은 기존의 약물보다 인체에 부작용이 적 고 치료 효과가 높아서 대체 약품으로 개발하려는 목적으로 조사되어 왔다 (Kim et al., 2011; Shihabudeen et al., 2011; Hyun et al., 2014). 특히, 식물에 존재하는 폴리페놀 화합물은 단백질과 결합하여 탄수화물 가수분해 효소인 α- glucosidase를 저해한다고 알려져 왔다 (Mai et al., 2007). 그러므로, 인체에 부작용이 없으면서도 당뇨병 치료에 우수한 효능을 가진 치료제 개발이 요구되고 있으며, 지금까지 빌레 나무 추출물의 α-glucosidase 저해 효과에 대한 과학적인 정 보는 보고된 바 없다. 이에, 빌레나무 잎과 가지의 항당뇨 효과를 검증하기 위하여, 메탄올 추출물과 그 분획물에 대한 α-glucosidase 저해 효과를 pNPG를 이용하여 측정하였다 (Table 1). 잎에서는 부탄올 분획물 (16.0 ± 1.1µg/ml)에서 저 해 효과를 나타내었으며 나머지 분획물에서는 낮은 저해 효 과를 보였다. 한편 가지에서는 에칠아세테이트 분획물 (0.8 ± 0.0µg/ml)에서 가장 높은 α-glucosidase 저해 효과를 보 였으며, 다음으로 부탄올 분획물 (3.8 ± 0.2µg/ml)에서 높은 저 해 효과를 보였다. 또한 메탄올 추출물 (22.3 ± 1.2µg/ml) 역 시 저해 효과를 보였다. 따라서, 빌레나무 가지 추출물이 전분 섭취에 의한 식후 혈당 상승을 억제시켜 줄 수 있는 잠재력이 있음을 확인하였다.

빌레나무의 페놀함량과 생리활성 (항산화 활성 및 항당뇨 활성) 간 상관관계가 있는지를 분석하기 위하여 피어슨 상관 계수를 사용하였다. Table 2는 페놀함량이 항산화 활성 및 α-glucosidase 저해 효과와 유의미함을 나타내었다. 페놀함량 은 프리라디칼 소거능 (r = −0.683, p < 0.01) 및 α-glucosidase 저해 효과 (r = −0.773, p < 0.001)와는 부의 상관관계를 나타냈 으며, 환원력 (r = 0.952, p < 0.001)과는 정의 상관관계를 나타 내었다. 또한 α-glucosidase 저해 효과는 프리라디칼 소거능력 (r = 0.893, p < 0.001)과 양의 상관관계를 나타내었으며, 환원력 (r = −0.76, p < 0.001)과는 음의 상관관계를 나타내었다. 이러한 연구 결과는 빌레나무 추출물의 페놀 화합물이 항산화 활성 및 α-glucosidase 저해 효과에 중요하게 기여하고 있음을 나타 낸다.

3. 항미생물 활성 측정

미생물 (박테리아, 곰팡이, 기생충 및 바이러스의 총칭)은 전 염성 질환의 원인이 되고 있으며, 이러한 질병을 이겨내기 위 하여 페니실린이나 스트렙토마이신과 같은 150개 이상의 항균 제들이 개발되었다 (World Health Organization, 2014). 하지 만 이러한 항생제에 대한 내성이 생기는 사례가 발생함에 따 라 천연물에서 미생물 저해 능력을 갖는 성분을 찾으려는 연 구가 활발히 진행되고 있다 (Jeong et al., 2010; Kim et al., 2010; Hyun et al., 2014). 본 실험에서는 심각한 식중독이나 감염의 원인이 되는 미생물에 대한 빌레나무 잎과 가지 추출 물의 항미생물 활성이 어느 정도 인지를 조사하기 위하여 실 시하였다 (Table 3). 그람 음성균 및 양성균을 대상으로 실험 을 한 결과, 잎에서는 활성이 약하거나 나타나지 않았다. 그러 나 가지의 에칠아세테이트 분획물에서는 Bacillus atrophaeusBacillus subtilis subsp. Spizizenii에서 250µg/ml의 농도에서 저해활성을 나타냈으며, Micrococcus luteusStaphylococcus epidermidis에서는 500µg/ml의 농도에서 저해활성을 나타냈다. 전체적으로 낮은 활성이었으며 잎보다는 가지에서 항미생물 활 성이 높음을 확인할 수 있었다. 또한 빌레나무 추출물은 그람 음성균보다 그람 양성균에 대하여 우수한 항균 활성을 나타냄 을 알 수 있었다.
