ABSTRACT

This study was carried out in order to investigate the photosynthesis response and leaf characteristics of Peucedanum japonicum growing in forest farming. The experiment was performed by leaf mold (pine tree and chestnut tree) and shading levels (0%, 35%, 50% and 75% shading). Light relative intensity was 100% (full sunlight), 60.3% (35% shading), 35.1% (50% shading), and 17.4% (75% shading) respectively. Light response curves of pine-leaf mold and chestnutleaf mold were the highest in control (full sunlight) and these were getting lower in the higher shading level. Photosynthesis capacity and light saturation point were indicated higher in chestnut-leaf mold within the same shading level. As the shading level increased, maximum photosynthesis rate decreased. And apparent quantum yield was not indicated statistically significant difference from all treatment. Leaf area, leaf length and leaf width were significant higher in 35% shading and control under chestnut-leaf mold in all treatment. As the shading level increased, LAR (leaf area ratio), SLA (specific leaf area) and SPAD value decreased in pine-leaf mold and chestnut-leaf mold. As a result of surveying the whole experiment, P. japonicum is judged better growth and higher yield by maintaining 35% shading (relative light intensity 60%) under chestnut-leaf mold in forest farming.

서 언

갯기름나물 (Peucedanum japonicum Thunberg)은 뿌리를 약용으로 이용하는 중요한 생약재로 생약규격집에 수재되어 있다.

우리나라에서는 갯기름나물 (식방풍)이 주로 재배, 생산되어 방풍으로 유통되고 있으며, 중국 원산지 방풍과는 식물 기원 이 다르다 (Gwak et al., 2011). 방풍류 생약에는 방풍 (진 방풍, 당방풍; Saposhnikovia divaricate (Turcz.) Schischk. = Ledebouriella seseloides (Hoffm.) H. Wolff), 갯기름나물 (식방풍, 빈해전호; Peucedanum japonicum Thunb.), 그리고 갯방풍 (해방풍, 빈방풍, 북사삼; Glehnia littoralis F. Schmidt ex Miq.) 등이 있다 (Chea et al., 2007).

한편, 자생지와 재배지에서 생육된 일부 산채산약초 등의 경 우에는 광합성 반응 조사를 통한 환경 및 생육 조건에 따른 식물의 건전도 평가에 관한 연구 결과가 보고되기도 하였다 (Kwon et al., 2009; Lee, 2002; Lee and Won, 2007; Lee et al., 2012a, b, c, d, 2014; Oh et al., 2010; Won et al., 2008). 하지만, 갯기름나물은 항암 활성 및 두통, 발한, 해열, 진통, 면역 등 성분 분석에 관한 연구가 다수 보고되었으나 (Kim, 2008; Lee et al., 2004; Shin et al., 1992; Son et al., 2013; Zheng et al., 2005) 자생지나 재배지의 광합성 반 응에 관한 연구는 전무한 실정이다.

따라서 본 연구에서는 임간재배지의 침엽 및 활엽 부엽토에 서 생육된 갯기름나물의 광합성 반응과 엽생장을 조사하여 갯 기름나물의 인위적 생산의 기초자료를 축척하고자 하였으며, 이러한 결과를 통해 보다 안전하게 생육할 수 있는 환경조건 을 구명하고 임간재배지 내에서 생산성을 높일 수 있는 기술 개발에 이바지하고자 하였다.


재료 및 방법
1. 공시식물 및 실험장소

본 실험의 공시식물은 갯기름나물 (Peucedanum japonicum Thunberg)이며, 갯기름나물의 유묘를 직접 생산하여 2014년 5월 8일에 각 처리구에 이식하였다. 실험은 국립산림과학원 남 부산림자원연구소의 진주시험림 내 임간재배 시험지에서 실시 하였으며, 시험지의 온도와 습도를 알아보기 위하여 2014년 5월 1일부터 8월 31일까지 온습도 측정기 (HOBO U23-001, ONSET Co. Corp., Santa Clara, CA, USA)를 지상으로부터 30cm 높이에 설치하였다 (Fig. 1).Table 1

2. 부엽토 및 차광 처리

침엽부엽토와 활엽부엽토가 갯기름나물의 광합성과 엽생장 에 미치는 영향 알아보기 위해 진주시험림 내에서 침엽부엽토 와 활엽부엽토를 각각 소나무 임분과 밤나무 임분에서 낙엽을 걷어내고 육안으로 균일한 입자 크기의 부엽토를 직접 채취하 여 처리구에 평당 10kg씩 처리하였다. 본 실험에 사용된 각 부엽토와 우리나라 산림토양의 이화학적 특성 비교는 Table 2 와 같다 (Jeong et al., 2002). 차광은 각 부엽토 내에 35%, 50%, 75%의 차광망을 이용하여 무차광을 포함한 총 4수준으 로 처리하였다 (Table 3).

3. 광합성 조사

부엽토 및 차광처리에 따른 갯기름나물의 광합성 조사를 위 해 2014년 8월 18일부터 5일 동안 건전한 잎을 대상으로 휴 대용 광합성 측정기 (Portable Photosynthesis System, LI- 6400, LI-COR Inc., Lincoln, NB, USA)를 이용하여 측정하 였다. 광도 PPFD (Photosynthetic Photon Flux Density)는 0, 100, 300, 600, 1000, 1500, 2000 μmol• m–2s–1의 총 7단 계를 각 처리구당 4반복으로 측정하였고, 광합성측정기의 leaf chamber에 유입되는 공기의 유량은 500 μmol• s–1, Chamber 온도는 25°C, CO2 농도는 400 μmol• mol–1, 상대습도는 60 ~ 70%로 조절하였다 (Kim et al., 2001). 이를 이용하여 광-광합성곡선 (light response curve)을 작성하고, Kok 효과 (Kok, 1948; Sharp et al., 1984)가 비교적 작게 나타나는 PPFD 0 ~ 100 μmol• m–2s–1 이하의 광도에서 광도와 광합성의 직선적 회귀식 (y = a + bx)을 구해 Kim과 Lee (2001)의 방 법으로 암호흡 (dark respiration rate), 광보상점 (light compensation point), 광포화점 (light saturation point), 최대 광합성율 (maximum photosynthesis rate), 순양자수율 (apparent quantum yield)을 각각 산출하였다.

4. 엽생장 및 엽록소 함량 조사

갯기름나물의 상대광도별 엽생장 특성을 분석하고자 생장조 사 실시 후 휴대용 엽면적 측정기인 Portable Area Meter (LI-3000C, LI-COR, Inc., Lincoln, NB, USA)를 이용하여 엽면적을 측정하였으며, L/W ratio (Leaf length/Leaf width), 엽면적비 (specific leaf area = Leaf area / Leaf dry weight, SLA), 엽면적율 (leaf area ratio = Leaf area / Total dry weight, LAR)을 구하였다.

엽록소 함량은 잎조직을 파괴하지 않고 반복측정이 가능한 Chlorophyll meter (SPAD-502, Konica Minolta Inc., Osaka, Japan)를 이용하여 10반복으로 측정하였다 (Woo et al., 2004).

5. 통계분석

부엽토 및 차광처리별 각각의 결과 값에 대한 분석은 SPSS version 20을 이용하여 분산분석 (ANOVA)을 실시하였으며, 통계적으로 차이가 유의한 경우 Duncan's Multiple Range Test (DMRT)를 실시하였다.


결과 및 고찰
1. 광합성 반응

침엽부엽토와 활엽부엽토에서 생육된 갯기름나물의 차광별 광-광합성곡선은 Fig. 2와 같으며, 이를 통해 암호흡, 광보상점, 광포화점, 최대광합성율, 순양자수율 등을 산출하였다.

침엽부엽토와 활엽부엽토에서 생육된 갯기름나물의 광도별 광합성능력의 경우, 침엽부엽토와 활엽부엽토 모두 전광에서 가장 높았으며, 그 다음은 각 부엽토에서 35%, 50%, 75% 차광 순으로 높았다. 전체적으로 가장 높은 광합성능력을 보 인 PPFD 2000 μmol• m–2s–1를 중심으로 비교할 때 활엽부엽토 의 전광, 35% 차광, 침엽부엽토의 전광, 활엽부엽토의 50%, 75% 차광 순으로 높았는데 이중에서 광합성능력이 가장 높은 활엽부엽토의 전광은 침엽부엽토의 전광 보다 약 1.5배 높았 으며, 가장 낮은 침엽부엽토의 75% 차광 보다 약 2.8배 높게 나타났다 (Fig. 2). 이러한 결과는 활엽부엽토가 갯기름나물의 광합성능력에 큰 영향을 미쳤기 때문으로 판단되며, 이는 유 효인산 등의 양분이 침엽부엽토 보다 활엽부엽토에서 많아 상 대적으로 공급이 원활했기 때문으로 사료된다 (Cho et al., 2010). 또한, 차광율이 높아질수록 갯기름나물의 광합성능력이 감소되는 것은 온도, 습도 등의 환경인자에 영향을 받는 임간 재배지 특성상 갯기름나물이 적정 환경에서 생육되지 못하여 광합성 기능이 저하되었기 때문으로 사료된다 (Kim et al., 2010).

한편, Kwon 등 (2009)은 이식 후 차광하여 생육한 산마늘, 곰취, 곤달비의 광합성능력의 경우, 전광에서 산마늘 (6.0 μmolCO2 • m–2s–1)은 가장 낮고 곰취 (11.8 μmolCO2 • m–2s–1) 와 곤달비 (12.2 μmolCO2 • m–2s–1)는 가장 높은 것으로 보고하 였는데, 본 실험의 갯기름나물의 경우에는 전광에서 가장 높 은 광합성능력을 보인 곰취, 곤달비와 유사한 결과를 보여 산 마늘 보다 내음성이 약한 식물로 판단되었다. 이러한 결과는 식물마다 광포화점이 다르기 때문에 나타난 결과로 사료된다 (Gardner et al., 1985).

암호흡은 전체가 2.0 ~ 4.6 μmol• m–2s–1로 처리구간 큰 차이를 보이지 않았다. 광보상점의 경우, 침엽부엽토에서는 58.9 ~ 64.3 μmol• m–2s–1로, 활엽부엽토에서는 36.4 ~ 65.5 μmol• m–2s–1 로 각각 조사되었는데 이들은 모든 처리구에서 유의적 차이를 보이지 않았다 (Table 4). 하지만, 활엽부엽토에서는 암호흡의 감소함에 따라 광보상점도 감소하여 정의 상관관계를 보이는 것으로 나타났다. 한편, 침엽부엽토 내 75% 차광에서 광보상 점이 높게 조사되었는데 이는 실험의 오차로 사료되며, 차후 의 실험에서는 보다 세심한 측정과 분석이 필요할 것으로 판 단된다.

광포화점과 최대 광합성율은 각 차광별로 비교할 경우에 모 두 침엽부엽토 보다 활엽부엽토에서 유의적 차이를 보이며 높 은 것으로 나타났다. 특히, 최대 광합성율의 경우에는 차광율 이 높아질수록 낮아지는 경향을 보였는데 이 또한 본 실험의 갯기름나물이 상대적으로 높은 광 환경에서 생육이 잘 되는 식물이기 때문에 나타난 결과로 사료된다.

약광 조건 (0 ~ 100 μmol• m–2s–1)에서의 광합성능력을 나타내 는 지표인 순양자수율은 빛 에너지를 화학에너지로 변환시키 는 광화학계의 활성을 반영한다 (Evans, 1987). 본 실험에서 상대적으로 광합성능력이 높은 활엽부엽토를 살펴볼 때 전광 에서 순양자수율이 높고 차광율이 높아질수록 광보상점, 광포 화점, 최대광합성율과 마찬가지로 감소하는 것을 알 수 있었다.

수분이용효율은 광합성 동화산물에 대한 수분손실율로서 광 합성의 감소로 인해 수분 이용이 제한될 때 즉각적인 수분이 용효율의 변화를 통해 건조에 대항한다고 한다 (Heschel et al., 2002). 본 실험에서 광도별 수분이용효율의 변화를 살펴보 면, 침엽부엽토와 활엽부엽토의 모든 처리구에서 광합성속도 가 양 (+)의 값으로 전환되는 광보상점 부근에서 수분이용효 율이 가장 낮았다. 그 중 침엽부엽토의 차광처리구 경우에는 광도가 상승함에 따라 수분이용효율 증가속도가 침엽부엽토의 전광과 활엽부엽토의 모든 처리구 보다 완만하게 이루어져 낮 은 수분이용효율을 보이는 것으로 나타났다. 그리고 대부분의 처리구에서는 광포화점 이상의 광도영역에서 수분이용효율이 완만하게 낮아지는 경향을 보였다 (Fig. 3).

광합성능력이 가장 높은 PPFD 2000 μmol• m–2s–1를 중심으 로 비교하면, 갯기름나물은 침엽부엽토와 활엽부엽토는 모두 35% 차광 이하에서 상대적으로 높은 수분이용효율을 보였다. 이러한 결과는 음지식물인 병풍쌈의 6월과 7월의 수분이용효 율이 차광율이 높아질수록 낮아진다는 연구보고와는 다른 경 향을 보였다 (Lee et al., 2012b). 한편, 전광을 제외한 차광 의 경우에는 침엽부엽토보다 활엽부엽토에서 더 높은 수분이 용효율을 보이는 것으로 나타났다.

2. 엽생장 특성

엽면적의 경우, 침엽부엽토에서 생육된 갯기름나물은 25.7 ~ 31.5cm2, 활엽부엽토에서 생육된 갯기름나물은 31.3 ~ 63.8cm2 로 침엽부엽토와 활엽부엽토 모두 35% 차광에서 가장 컸으 며, 차광별로는 활엽부엽토에서 생육된 갯기름나물이 더 큰 것 으로 나타났다 (Table 5). 전체적으로는 활엽부엽토의 35% 차 광과 전광에서 유의적으로 크게 나타났으며, 차광별로는 모든 차광에서 활엽부엽토가 침엽부엽토 보다 1.2 ~ 2.0배 더 컸는데 이는 유효성분이 더 많은 활엽부엽토의 영향으로 판단된다. 일 반적으로 낮은 광조건에서 생장한 잎의 면적이 넓은 것으로 알려져 있고 (Salisbury and Ross, 1992), 병풍쌈 (Yoon et al., 2014)과 수리취 (Lee et al., 2012c)의 경우에는 이와 동 일한 결과를 보였으나 본 실험의 갯기름나물은 다른 경향을 보이는 것으로 나타났다. 이는 본 실험의 시험지가 다양한 환 경인자에 영향을 받는 임간에 위치하였기 때문으로 사료된다.

엽장과 엽폭도 침엽부엽토와 활엽부엽토 모두 35% 차광에 서 가장 컸으며, 전체적으로는 활엽부엽토의 35% 차광과 전 광에서 유의적 차이를 보이며 각각 10.2cm, 6.4cm와 9.5cm, 6.2cm로 크게 나타났다. L/W는 엽장과 엽폭이 유의적으로 큰 활엽부엽토의 전광과 35% 차광에서 가장 작았는데 이는 엽폭 이 상대적으로 다른 처리구 보다 1.4 ~ 1.7배 컸기 때문에 나 타난 결과로 사료된다.

엽면적 비율을 나타내는 LAR (leaf area ratio)은 침엽부엽 토와 활엽부엽토 모두 차광율이 높아질수록 커져 75% 차광에 서 53.5cm2• g–1, 45.0cm2• g–1로 가장 높았다. 또한, 엽두께를 간접적으로 나타내는 SLA (specific leaf area)는 이 값이 클 수록 엽두께가 얇은 것을 의미하는데, 본 실험에서는 침엽부 엽토와 활엽부엽토 모두 차광율이 높아질수록 커져 75% 차광에서 각각 165.3cm2• g–1, 152.1cm2• g–1로 가장 높았다 (Table 5). 이는 차광율이 높아질수록 갯기름나물의 엽두께가 얇아진다는 것을 의미한다.

한편, 약용으로 이용이 가능한 다년생 산채인 병풍쌈 (Lee et al., 2012b), 고려엉겅퀴 (Lee et al., 2012d), 수리취 (Lee et al., 2012c)도 차광율이 높아질수록 SLA (specific leaf area)가 감소하는 것으로 보고된 바 있어 식물에 따라 다른 것 을 알 수 있다.

SPAD를 이용해 엽록소 함량을 조사한 결과, 본 실험에서 침엽부엽토와 활엽부엽토에서 생육된 갯기름나물 모두 차광율 이 높아질수록 높은 엽록소 함량을 보였는데 이는 유입되는 광량이 낮은 환경에서 생육되어 나타난 결과로 판단된다 (Hansen et al., 2002; Viji et al., 1997). 또한, 이러한 결과 는 부족한 광 환경에서 광합성 효율을 증대시키기 위한 생리 적 반응으로 사료되며, 병풍쌈 (Lee et al., 2012b)과 수리취 (Lee et al., 2012c) 뿐만 아니라 산마늘, 곰취, 곤달비 (Kwon et al., 2009)와도 동일한 경향을 보이는 것으로 나타 났다.

임간재배지에서 갯기름나물의 생육은 침엽부엽토 보다 활엽 부엽토가 많은 장소에서 35% 차광 (상대광도 60%) 이하의 수준을 유지하는 것이 고품질 갯기름나물의 생산 및 생산량 증대에 보다 효과적일 것으로 판단된다.
